Zebrafish and neonatal mice have become powerful animal models for regenerative studies because of their regenerative capacity after injury.Understanding the cellular and molecular mechanisms on their role in heart regeneration holds great potential for priming adult mammalian heart for regeneration. By establishing a chemical screening, we have identified a small-molecule cocktail 5SM that enables mammalian cardiomyocyte proliferation and heart regeneration (Cell Stem Cell 2022; J Mol Cell Biol 2023). Ongoing studies focus on determining the underlying mechanisms by multiomics approaches, as well as searching for single chemical molecule that sufficiently promotes heart regeneration in small and large animal models. I will present data how 5SM act on several heart cell types to coordinate heart regeneration in mice after myocardial infarction, and thus our works may shed light on both fundamental mechanisms on heart regeneration and translational implication on heart disease.
